Cell Lysis (Total Protein Extraction Reagents) Extract total protein from mammalian tissue or cell samples using mild but efficient lysis reagents. These reagents have been optimized and validated with specific tissue types, as well as in primary and cultured mammalian cells. Detergents for Protein Solubilization Detergents are amphipathic molecules containing both a nonpolar “tail” having aliphatic or aromatic character, and a polar “head.” The ionic character of the polar head group forms the basis for broad classification of detergents; they may be ionic (charged, either anionic or cationic), nonionic (uncharged) or zwitterionic (having both positively and negatively charged groups, but with a net charge of zero). Detergents are frequently used in cell lysis reagent formulation, for protein solubilization procedures, in wash buffers for ELISA, and for other protein research methods. Thermo Scientific™ Pierce Surfact-Amps detergents are highly purified, precisely diluted (10%) formulations that are ideal for applications or assays that are sensitive to contaminants that are present in unpurified detergents. Pierce Coomassie (Bradford) Assay The Pierce Coomassie (Bradford) Protein Assay Kit is a ready-to-use, stable formulation of the traditional Bradford assay reagent for measurement of total protein concentration compared to a protein standard. The kit includes Coomassie Protein Assay Reagent and a package of Albumin Standard ampules. The Pierce Coomassie Protein Assay Kit is a ready-to-use formulation of the popular assay reagent originally described by Bradford in 1976. When mixed with a protein solution, the acidic Coomassie-dye reagent changes color from brown to blue in proportion to the amount of protein present in the sample. Protein determinations are made by comparison to the color response of protein assay standards, usually prepared as a series of known dilutions of bovine serum albumin (BSA) or bovine gamma globulin (BGG). Pierce 600nm Protein Assay The Pierce 660-nm Protein Assay is a ready-to-use, detergent- and reducing agent-compatible assay reagent to quickly measure total protein concentration compared to a protein standard. The assay is more linear than Coomassie-based Bradford assays and compatible with higher concentrations of most detergents, reducing agents, and other commonly used reagents. The Pierce 660nm Protein Assay is a quick, ready-to-use colorimetric method for measuring protein concentration. The assay is more linear than coomassie-based Bradford assays and compatible with higher concentrations of most detergents, reducing agents and other commonly used reagents. The accessory Ionic Detergent Compatibility Reagent (IDCR) provides for even broader detergent compatibility, making the Pierce 660nm Protein Assay the only protein assay that is suitable for samples containing Laemmli SDS sample buffer with bromophenol blue. SuperSignal Protein Ladder Thermo Scientific™ SuperSignal Molecular Weight Protein Ladders (20 to 150 kDa) are a ready-to-use mix of recombinant proteins with IgG-binding sites that provide reliable and proportional band intensities in stained gels and immunoblots developed with chemiluminescent, fluorescent, chromogenic or other detection systems. The SuperSignal Molecular Weight Protein Ladders and SuperSignal Enhanced Molecular Weight Protein Ladders contain a ready-to-use stabilized mixture of eight recombinant proteins ranging in size from 20 to 150 kDa. These recombinant proteins bind antibodies used in the Western blot through an IgG binding site. The protein markers can then be visualized either using appropriate substrates for enzyme-labeled antibodies or via fluorescent dye-labeled antibodies. Pierce Silver Stain Thermo Scientific™ Pierce Silver Stain Kit is a rapid, ultra-sensitive and versatile silver stain system for protein detection in polyacrylamide gels, yielding consistent and reliable results with many 1D and 2D gel types for both first-time and experienced users. Krypton Fluorescent Protein Stain Thermo Scientific™ Pierce Krypton Fluorescent Protein Stain enables sensitive fluorescent visualization (520 nm excitation; 580 nm emission) of proteins separated by polyacrylamide gel electrophoresis or on transfer membranes using popular commercial fluorescence imagers. This fluorescent protein gel stain is for bright yellow-orange fluorescent detection of proteins separated by 1D or 2D polyacrylamide gel electrophoresis (SDS-PAGE). Krypton Stain uses a protein-binding dye with fluorescence properties (Ex/Em = 520/580 nm) suitable for visualization with a variety of standard laser-based and filter-based fluorescence imagers and documentation systems. Proteins are detectable at concentrations greater than 0.25 ng per band in a typical mini-gel lane (greater than 2 ng for the rapid staining protocol). Avidin, streptavidin, and neutravidin bind very strongly and specifically to biotin. However, each protein has its limitations in certain assays. Avidin is glycosylated which may lead to nonspecific lectin binding in assays. Streptavidin contains an RYD motif, a bacterial recognition sequence, which can cause background binding with certain samples. Another alternative is to use neutravidin protein which is an exclusive, deglycosylated form of Avidin that avoids the drawbacks of both native avidin and streptavidin. ELISA Enzyme Substrates
Colorimetric substrates Find a variety of colorimetric (chromogenic) substrates for ELISA applications for horseradish peroxidase (HRP) and for alkaline phosphatase (AP) to meet your sensitivity and experimental needs. Colorimetric ELISA substrates at a glance TMB (3,3′,5,5′-tetramethylbenzidine) soluble substrates yield a blue color when detecting HRP. The major absorbance maxima or the reaction product are 370 nm and 652 nm. The color then changes to yellow with the addition of sulfuric or phosphoric acid (stop solution) with maximum absorbance at 450 nm. TMB is very sensitive and may produce significant background signal if too much protein or antibody is used. TMB is more quickly oxidized than other HRP substrates, resulting in faster color development. ABTS (2,2′-Azinobis [3-ethylbenzothiazoline-6-sulfonic acid]-diammonium salt) is used to detect HRP and yields a water-soluble green end reaction product. The green product has two major absorbance peaks – 410 nm and 650 nm. ABTS is less sensitive than the OPD and TMB substrates for HRP detection. Color development is slow (approximately 20 minutes) which may be advantageous if unacceptable background results from the use of the OPD or TMB substrates due to higher sensitivities. ABTS is available in either tablet or a ready-to-use for formulation. OPD (o-phenylenediamine dihydrochloride) is used to detect HRP and yields a water soluble yellow-orange reaction product. The reaction product has an absorbance maximum of 492 nm. Pierce LAL Chromogenic Endotoxin Quantitation Kit The Thermo Scientific™ Pierce LAL Chromogenic Endotoxin Quantitation Kit measures the amount of endotoxin in a protein, peptide or antibody sample using the Limulus Amebocyte Lysate (LAL) assay. The endotoxin concentration in a sample is measured using the Pierce LAL Chromogenic Endotoxin Quantitation Kit via a chromogenic signal generated in the presence of endotoxins. Samples can be measured on a microplate absorbance reader at 405 nm. A standard curve is created using the E. coli endotoxin standard included with each kit to calculate endotoxin levels as low as 0.1 EU/mL. Pierce Glycoprotein Carbohydrate Estimation Kit Thermo Scientific™ Pierce Glycoprotein Carbohydrate Estimation Kit enables the amount of protein glycosylation to be measured as the percent of total purified protein mass. The Glycoprotein Carbohydrate Estimation Kit includes six purified glycoprotein standards and the required assay reagents to effectively estimate the amount of oxidizable glycosylation (% carbohydrate by weight) of purified protein samples. Sugar groups in the glycoprotein sample are first oxidized with sodium meta-periodate to produce detectable aldehyde groups. Then the sample is reacted with the colorimetric Glycoprotein Detection Reagent. Absorbance at 550 nm of the resulting purple reaction product is measured with a spectrophotometer or plate reader. Pierce Quantitative Colorimetric Peptide Assay The Pierce Quantitative Colorimetric Peptide Assay provides modified BCA reagents for the reduction of Cu+2 to Cu+1 and a proprietary chelator that have been optimized for the quantitation of peptide mixtures. In this reaction, the copper is first reduced by the amide backbone of peptides under alkaline conditions (Biuret reaction). The proprietary chelator then couples with the reduced copper to form a bright red complex with absorbance of 480nm. The signal produced from this reaction is 3-4 fold more sensitive than the Micro-BCA Protein Assay for peptide analysis. The kit also contains a high quality peptide digest reference standard for use in generating linear standard curves and to serve as a calibration control. This colorimetric peptide assay requires a small amount of sample (20μL) and has a working peptide concentration range of 25 to 1000μg/mL. The increased sensitivity, low sample assay volume, and included reference standard are essential for accurate and robust measurement of peptide digest samples, especially for mass spectrometry applications. | |

There are several strategies that can be deployed to increase target protein identifications: utilize post-translational modification (PTM) specific affinity binding, such as phosphopeptide enrichment, or through fractionation, such as high pH reversed-phase chromatography which separates peptides by hydrophobicity. The novel Thermo Scientific™ ActivX™ probes and enrichment kits enable the specific targeting of several enzyme classes, including kinases, GTPases, and serine hydrolases, which are covalently labeled in the active site at the protein level and enriched after digestion.

Pierce Peptide Clean-up Solutions After isolation of peptides, salts and buffers can be removed using reversed phase (RP) resins, of which the C18 matrix is the most ideal for the capture of hydrophobic peptides. The peptides bind to reverse-phase columns in high-aqueous mobile phase, salts and buffers are washed off, and the peptides are eluted using a high-organic mobile phase. However, hydophilic peptides, including phosphopeptides, may bind to C18 resins poorly, so graphite spin columns are recommended for better peptide recovery. However, C18 and graphite resins do not efficiently remove detergents, which can contaminate instruments and interfere with column binding, elution and ionization. The Thermo Scientific™ Detergent Removal products efficiently bind to and remove high concentrations of a wide variety of detergents that are commonly used during sample processing.
