Cell Lysis (Total Protein Extraction Reagents)

Extract total protein from mammalian tissue or cell samples using mild but efficient lysis reagents. These reagents have been optimized and validated with specific tissue types, as well as in primary and cultured mammalian cells. Protein extracts are compatible with most protein assays and typical downstream applications. These reagents offer higher protein yield than other commercially available or homemade formulations. The convenient single formulation reagents typically allow samples to be processed in 5–10 minutes. When lysing primary and cultured cells, mechanical disruption is not required.

Detergents for Protein Solubilization

Detergents are amphipathic molecules containing both a nonpolar “tail” having aliphatic or aromatic character, and a polar “head.” The ionic character of the polar head group forms the basis for broad classification of detergents; they may be ionic (charged, either anionic or cationic), nonionic (uncharged) or zwitterionic (having both positively and negatively charged groups, but with a net charge of zero).

Detergents are frequently used in cell lysis reagent formulation, for protein solubilization procedures, in wash buffers for ELISA, and for other protein research methods.

Thermo Scientific™ Pierce Surfact-Amps detergents are highly purified, precisely diluted (10%) formulations that are ideal for applications or assays that are sensitive to contaminants that are present in unpurified detergents. Protease and Phosphatase Inhibitors

Protease and phosphatase inhibitors are essential components of most cell lysis and protein extraction procedures. These inhibitors block or inactivate endogenous proteolytic and phospholytic enzymes that are released from subcellular compartments during cell lysis and would otherwise degrade proteins of interest and their activation states.

Dialysis, Desalting and Concentrators

After cell lysis or isolation, lysates often contain contaminants that are not compatible with downstream steps. We offer a variety of specialty devices and resins that simply and efficiently desalt, exchange buffers, and remove detergents from samples. In addition, if the protein concentration is too dilute for further processing or analysis, the sample can be concentrated quickly using centrifugal concentrators.

Pierce BCA Protein Assay

Pierce BCA Protein Assay Kit is a two-component, high-precision, detergent-compatible protein assay for determination of protein concentration. Pierce BCA reagents provide accurate determination of protein concentration with most sample types encountered in protein research. The Pierce BCA assay can be used to assess yields in whole cell lysates, affinity-column fractions, purified protein samples, as well as to monitor protein contamination in industrial applications. Compared to most dye-binding methods, the BCA assay is affected much less by protein compositional differences, providing greater concentration accuracy.

Pierce Coomassie (Bradford) Assay

The Pierce Coomassie (Bradford) Protein Assay Kit is a ready-to-use, stable formulation of the traditional Bradford assay reagent for measurement of total protein concentration compared to a protein standard. The kit includes Coomassie Protein Assay Reagent and a package of Albumin Standard ampules.

The Pierce Coomassie Protein Assay Kit is a ready-to-use formulation of the popular assay reagent originally described by Bradford in 1976. When mixed with a protein solution, the acidic Coomassie-dye reagent changes color from brown to blue in proportion to the amount of protein present in the sample. Protein determinations are made by comparison to the color response of protein assay standards, usually prepared as a series of known dilutions of bovine serum albumin (BSA) or bovine gamma globulin (BGG). The simple procedure is adaptable to nearly any volume scale, including test tubes, cuvettes, and microplates. The protein assay is compatible with most salts, solvents, buffers, thiols, reducing substances, and metal chelating agents encountered in protein samples.

Pierce Modified Lowry Protein Assay

The Pierce Modified Lowry Protein Assay is a stable form of a traditional, two-component, folin phenol- and copper-based reagent system to measure total protein concentration compared to a protein standard.

Features include:

Pierce 600nm Protein Assay

The Pierce 660-nm Protein Assay is a ready-to-use, detergent- and reducing agent-compatible assay reagent to quickly measure total protein concentration compared to a protein standard. The assay is more linear than Coomassie-based Bradford assays and compatible with higher concentrations of most detergents, reducing agents, and other commonly used reagents.

The Pierce 660nm Protein Assay is a quick, ready-to-use colorimetric method for measuring protein concentration. The assay is more linear than coomassie-based Bradford assays and compatible with higher concentrations of most detergents, reducing agents and other commonly used reagents. The accessory Ionic Detergent Compatibility Reagent (IDCR) provides for even broader detergent compatibility, making the Pierce 660nm Protein Assay the only protein assay that is suitable for samples containing Laemmli SDS sample buffer with bromophenol blue.

Although the Pierce 660nm Protein Assay produces a higher level of protein-to-protein variation (37%) than other assays, such as the BCA Protein Assay, the simpler single-reagent format and broader substance compatibility make the 660nm Assay more convenient for many routine applications. The Pierce 660nm Protein Assay can be performed in either a test tube or microplate format.

The assay is based on a proprietary dye-metal complex that binds to proteins in an acidic medium. Upon binding, the reddish dye-metal complex turns to green, resulting in an absorbance shift that is measurable at wavelengths near 660nm (645 to 670). The dye interacts mainly with basic residues in proteins, such as histidine, arginine and lysine and to a lesser extent tyrosine, tryptophan and phenylalanine.

This prestained protein MW marker is designed for monitoring the progress of SDS-polyacrylamide gel electrophoresis, for assessing transfer efficiency onto PVDF, nylon and nitrocellulose membranes, and for estimating the approximate size of separated proteins that have been made visible with gel stains or Western blot detection reagents.

PageRuler Protein Ladder

Thermo Scientific™ PageRuler Prestained Protein Ladder is a mixture of 10 blue-, orange-, and green-stained proteins (10 to 180 kDa) or 9 blue-, orange- and green-stained proteins (10 to 250 kDa) for use as size standards in protein electrophoresis (SDS-PAGE) and western blotting. The protein ladder is supplied in a ready-to-use format for direct loading onto gels; no need to heat, reduce, or add sample buffer prior to use.

Spectra Protein Ladder

Thermo Scientific™ Spectra Multicolor Broad Range Protein Ladder is a 4-color protein standard containing 10 prestained proteins (10 to 260kDa) for use as size standards in gel electrophoresis and Western blotting.

This prestained protein MW marker is designed for monitoring the progress of SDS-polyacrylamide gel electrophoresis, for assessing transfer efficiency onto PVDF, nylon and nitrocellulose membranes, and for estimating the approximate size of separated proteins that have been made visible with gel stains or Western blot detection reagents. Four different chromophores (blue, orange, green, pink) are bound to the different component proteins, producing a brightly colored ladder with an easy-to-remember pattern.

SuperSignal Protein Ladder

Thermo Scientific™ SuperSignal Molecular Weight Protein Ladders (20 to 150 kDa) are a ready-to-use mix of recombinant proteins with IgG-binding sites that provide reliable and proportional band intensities in stained gels and immunoblots developed with chemiluminescent, fluorescent, chromogenic or other detection systems.

The SuperSignal Molecular Weight Protein Ladders and SuperSignal Enhanced Molecular Weight Protein Ladders contain a ready-to-use stabilized mixture of eight recombinant proteins ranging in size from 20 to 150 kDa. These recombinant proteins bind antibodies used in the Western blot through an IgG binding site. The protein markers can then be visualized either using appropriate substrates for enzyme-labeled antibodies or via fluorescent dye-labeled antibodies. The molecular weight marker mix also contains a special pink tracking dye to monitor the electrophoresis run and membrane transfer without the need for additional colored markers. The standards are compatible with nitrocellulose and PVDF membranes, multiple blocking buffers and many other detection methods.

We offer two versions of the standard. The regular formulation is appropriate for use with most rabbit and other non-mouse polyclonal antibodies. The enhanced standard is formulated specifically for applications requiring mouse monoclonal antibodies and experiments where very dilute antibody concentrations are used.

GelCode Protein Gel Stain

GelCode Blue Stain Reagent is a ready-to-use protein stain based on colloidal coomassie dye G-250 that provides nanogram-level detection in polyacrylamide gels and for in-blot detection.

Pierce Silver Stain

Thermo Scientific™ Pierce Silver Stain Kit is a rapid, ultra-sensitive and versatile silver stain system for protein detection in polyacrylamide gels, yielding consistent and reliable results with many 1D and 2D gel types for both first-time and experienced users.

The Pierce Silver Stain Kit is a metallic silver (Ag) protein stain that yields a remarkably clear and uniform gel background while delivering consistent, high-sensitivity staining results at each and every use. In standard mini gels, proteins are detectable at greater than 0.25ng per band or spot. The protocol has been optimized for flexibility by allowing short or overnight gel fixation and staining steps without affecting staining performance (sensitivity or clarity). A short (one-minute) sensitization step, performed after gel fixation, yields results that are free of the characteristically dark or blotchy backgrounds often seen with homemade or other commercially available silver stains. This feature is beneficial when performing densitometric analysis of silver stained gels.

Krypton Fluorescent Protein Stain

Thermo Scientific™ Pierce Krypton Fluorescent Protein Stain enables sensitive fluorescent visualization (520 nm excitation; 580 nm emission) of proteins separated by polyacrylamide gel electrophoresis or on transfer membranes using popular commercial fluorescence imagers.

This fluorescent protein gel stain is for bright yellow-orange fluorescent detection of proteins separated by 1D or 2D polyacrylamide gel electrophoresis (SDS-PAGE). Krypton Stain uses a protein-binding dye with fluorescence properties (Ex/Em = 520/580 nm) suitable for visualization with a variety of standard laser-based and filter-based fluorescence imagers and documentation systems. Proteins are detectable at concentrations greater than 0.25 ng per band in a typical mini-gel lane (greater than 2 ng for the rapid staining protocol). Krypton Stain provides high signal intensity, exhibits minimal protein-to-protein variation, has an excellent quantitative dynamic range and is compatible with subsequent destaining and analysis by mass spectrometry.

Pierce Fast Western Blotting Kits offer optimized reagents that shorten the time to perform a typical western blot from 4h down to approximately 55 mins. These kits help reduce hands-on and overall blotting time using Thermo Scientific™ ECL and SuperSignal Chemiluminescent substrates.

We offer streptavidin, neutravidin and avidin protein conjugates for biotinylated antibody or protein probe detection including HRP, AP conjugates and high-intensity fluorescent and infrared conjugates. Avidin, streptavidin, and neutravidin bind very strongly and specifically to biotin. However, each protein has its limitations in certain assays. Avidin is glycosylated which may lead to nonspecific lectin binding in assays. Streptavidin contains an RYD motif, a bacterial recognition sequence, which can cause background binding with certain samples. ELISA Enzyme Substrates

Colorimetric substrates

Find a variety of colorimetric (chromogenic) substrates for ELISA applications for horseradish peroxidase (HRP) and for alkaline phosphatase (AP) to meet your sensitivity and experimental needs.

Colorimetric ELISA substrates at a glance

TMB (3,3′,5,5′-tetramethylbenzidine) soluble substrates yield a blue color when detecting HRP. The major absorbance maxima or the reaction product are 370 nm and 652 nm. The color then changes to yellow with the addition of sulfuric or phosphoric acid (stop solution) with maximum absorbance at 450 nm. TMB is very sensitive and may produce significant background signal if too much protein or antibody is used. TMB is more quickly oxidized than other HRP substrates, resulting in faster color development.

ABTS (2,2′-Azinobis [3-ethylbenzothiazoline-6-sulfonic acid]-diammonium salt) is used to detect HRP and yields a water-soluble green end reaction product. The green product has two major absorbance peaks – 410 nm and 650 nm. ABTS is less sensitive than the OPD and TMB substrates for HRP detection. Color development is slow (approximately 20 minutes) which may be advantageous if unacceptable background results from the use of the OPD or TMB substrates due to higher sensitivities. ABTS is available in either tablet or a ready-to-use for formulation.

OPD (o-phenylenediamine dihydrochloride) is used to detect HRP and yields a water soluble yellow-orange reaction product. The reaction product has an absorbance maximum of 492 nm. OPD is available in either powder or tablet forms and easily prepared by dissolving in Stable Peroxide Substrate Buffer or buffered hydrogen peroxide solution.

PNPP (p-Nitrophenyl Phosphate, Disodium Salt) is a widely used substrate for detecting alkaline phosphatase in ELISA applications. PNPP produces a yellow water-soluble reaction product that absorbs light at 405 nm. PNPP is available either as a crystalline powder, 5 mg tablets, or as a ready-to-use formulation.

SuperSignal ELISA Pico Chemiluminescent Substrate provides excellent performance for a large range of target protein amounts and is easily optimized to detect with greater sensitivity than entry-level colorimetric substrates. Rapid signal generation with 5–30 minute signal stability depending on HRP concentration.

SuperSignal ELISA Femto Maximum Sensitivity Substrate is one of the most sensitive substrates available for ELISA applications. When properly optimized, the lower detection limit is 1 to 10 orders of magnitude lower than commonly used colorimetric substrates. However, without proper optimization, it is easy to overwhelm the system with protein and enzyme, resulting in high background and possibly negative results.

QuantaBlu Fluorogenic Substrate has a large linear detection range with low-end linearity for detection of HRP. QuantaRed Enhanced Chemifluorescent Substrate is the most sensitive fluorescent ELISA substrate available for HRP detection. The fluorescent reaction product (resorufin) is stable for 4 hours with an Emax/Amax of 585 nm/570 nm when the reaction is stopped. The red-shifted resorufin reaction product permits detection at a wavelength with less interference from autofluorescence that can occur in biological samples.

Pierce Chromogenic Endotoxin Quant Kit

The Thermo Scientific™ Pierce Chromogenic Endotoxin Quant Kit is a highly sensitive endpoint assay that accurately measures and detects endotoxin (lipopolysaccharide) in a protein, peptide, nucleic acid, or antibody sample using the amebocyte lysate assay. The kit enables detection within two linear sensitivity ranges of 0.01–0.1 EU/mL and 0.1–1.0 EU/mL.

Pierce LAL Chromogenic Endotoxin Quantitation Kit

The Thermo Scientific™ Pierce LAL Chromogenic Endotoxin Quantitation Kit measures the amount of endotoxin in a protein, peptide or antibody sample using the Limulus Amebocyte Lysate (LAL) assay.

The endotoxin concentration in a sample is measured using the Pierce LAL Chromogenic Endotoxin Quantitation Kit via a chromogenic signal generated in the presence of endotoxins. Samples can be measured on a microplate absorbance reader at 405 nm. A standard curve is created using the E. coli endotoxin standard included with each kit to calculate endotoxin levels as low as 0.1 EU/mL. Protein and antibody samples can be assayed in as little as 30 minutes. Determining endotoxin levels is important to assess the efficiency of endotoxin removal methods and prevent endotoxic shock, inflammation and/or sepsis in tissue culture cells and animals injected with endotoxin-contaminated proteins.

Pierce Glycoprotein Carbohydrate Estimation Kit

Thermo Scientific™ Pierce Glycoprotein Carbohydrate Estimation Kit enables the amount of protein glycosylation to be measured as the percent of total purified protein mass.

The Glycoprotein Carbohydrate Estimation Kit includes six purified glycoprotein standards and the required assay reagents to effectively estimate the amount of oxidizable glycosylation (% carbohydrate by weight) of purified protein samples. Sugar groups in the glycoprotein sample are first oxidized with sodium meta-periodate to produce detectable aldehyde groups. Then the sample is reacted with the colorimetric Glycoprotein Detection Reagent. Absorbance at 550 nm of the resulting purple reaction product is measured with a spectrophotometer or plate reader. Finally, the carbohydrate content in the glycoprotein is calculated by comparison to results from the five glycoprotein standards that are included in the kit.

Tandem Mass Tag Labeling Reagents & Kits

The Thermo Scientific™ Tandem Mass Tag Reagents are designed to enable identification and quantitation of proteins in different samples using tandem mass spectrometry (MS). All mass tagging reagents within a set have the same nominal mass (i.e., are isobaric) and chemical structure composed of an amine-reactive NHS ester group, a spacer arm (mass normalizer), and a mass reporter.

The standard tandem mass tag (TMT) labeling kits and reagents enable multiplex (6-plex to 11-plex) relative quantitation using high-resolution MS for samples prepared from cells or tissues. The TMT 10plex and 11plex label reagents share an identical structure with TMTzero, TMTduplex, and TMT 6plex reagents but contain different numbers and combinations of 13C and 15N isotopes in the mass reporter.

Abundant Protein Depletion

Depletion is used to reduce the complexity of biological samples such as serum, plasma, or biofluids, which contain high concentrations of albumin and immunoglobulins. These products utilize immunoaffinity techniques to enable the removal of the most abundant proteins, thus enhancing the detection of lower-abundance proteins in both discovery and targeted proteomic analyses.

Pierce Quantitative Colorimetric Peptide Assay

The Pierce Quantitative Colorimetric Peptide Assay provides modified BCA reagents for the reduction of Cu+2 to Cu+1 and a proprietary chelator that have been optimized for the quantitation of peptide mixtures. In this reaction, the copper is first reduced by the amide backbone of peptides under alkaline conditions (Biuret reaction). The proprietary chelator then couples with the reduced copper to form a bright red complex with absorbance of 480nm. The signal produced from this reaction is 3-4 fold more sensitive than the Micro-BCA Protein Assay for peptide analysis.

The kit also contains a high quality peptide digest reference standard for use in generating linear standard curves and to serve as a calibration control. This colorimetric peptide assay requires a small amount of sample (20μL) and has a working peptide concentration range of 25 to 1000μg/mL. The increased sensitivity, low sample assay volume, and included reference standard are essential for accurate and robust measurement of peptide digest samples, especially for mass spectrometry applications.

Protein Digestion

Sequence specific proteases are utilized to cleave proteins into smaller fragments, or peptides. Trypsin is the protease of choice for protein digestion. However, separate or sequential digestion with alternative proteases, such as Glu-C, LysN, Lys-C, Asp-N or Chymotrypsin, can improve individual protein sequence coverage or generate unique peptide sequences for different MS applications. The Thermo Scientific™ Pierce Proteases are highly purified and modified for optimal protein digestion and validated for use in MS. The Thermo Scientific EasyPep Mini MS Sample Prep Kit provides pre-formulated reagents and a robust method for the preparation of high quality samples that are ready for MS analysis in less than 3 hours.

Peptide Enrichment and Fractionation

Post digestion, peptide enrichment and/or fractionation is often necessary to detect peptides from low-abundant proteins and/or for reducing sample complexity for proteomic analysis.

There are several strategies that can be deployed to increase target protein identifications: utilize post-translational modification (PTM) specific affinity binding, such as phosphopeptide enrichment, or through fractionation, such as high pH reversed-phase chromatography which separates peptides by hydrophobicity.

The novel Thermo Scientific™ ActivX™ probes and enrichment kits enable the specific targeting of several enzyme classes, including kinases, GTPases, and serine hydrolases, which are covalently labeled in the active site at the protein level and enriched after digestion.

Pierce Peptide Clean-up Solutions

After isolation of peptides, salts and buffers can be removed using reversed phase (RP) resins, of which the C18 matrix is the most ideal for the capture of hydrophobic peptides. The peptides bind to reverse-phase columns in high-aqueous mobile phase, salts and buffers are washed off, and the peptides are eluted using a high-organic mobile phase. However, hydophilic peptides, including phosphopeptides, may bind to C18 resins poorly, so graphite spin columns are recommended for better peptide recovery. However, C18 and graphite resins do not efficiently remove detergents, which can contaminate instruments and interfere with column binding, elution and ionization. The Thermo Scientific™ Detergent Removal products efficiently bind to and remove high concentrations of a wide variety of detergents that are commonly used during sample processing.
